Human Viral Isolate Reference Sheet

Catalogue Item Name: Ad 22-1

Classification:  Adenovirus serotype 22              Strain:  undetermined

Passage:  2   Culture Host:  A549 cells Time period Host cells infected: 4 days

Original Source Information: This adenovirus isolate was obtained from an Iowa virology laboratory.  It was cultured from bronchial specimens collected in March 2002 from a hospitalized 37–year-old male with pneumonia seen in Iowa. The patient died during the hospitalization.

Method of Identification Confirmation:  A hospital virology laboratory performed cell culture and adenovirus monoclonal fluorescent antibody staining confirmation.  The Emerging Pathogens Laboratory repeated cell culturing on A549 cells, for microneutralization serotyping using antisera 1-5, 7, and 21.  Results of the microneutralization testing were uninterpretable.  Restriction enzyme analysis, of the purified DNA with BamHi revealed a pattern differing from the Ad 7 strains given to us by CDC (Ad 7a, Ad 7b, Ad 7d/d2, Ad 7h, and Ad 7p). The isolate was given to the CDC for identification who determined it to be an adenovirus of species D and further serotyped it as adenovirus serotype 22.
