Original Source Information:  This specimen was collected in a clinical virology lab in Iowa in December 2002 from a nasal wash sample of a 5 year-old female.

 

Method of Identification Confirmation:  Before evaluating the nasal wash sample specimen for hMPV, it was found to be negative by routine viral culture for other common respiratory pathogens including: influenza A, influenza B, parainfluenza, adenovirus, and RSV.  The specimen was inoculated onto LLC-MK2 cells and incubated for a total of 21 days.  CPE was observed on 14 and 21 days.  The supernatant tested positive on days 7, 14, and 21 for hMPV by an RT-PCR procedure targeting F gene (347bp amplicon) and N gene (213bp amplicon) as published in JID 2003:187;785-90 and JCM Jan 2003:Vol.4,No.1;100-5 respectively.  The shell vials supernatant was harvested, aliquotted, and frozen at -70ºC on days 7, 14, and 21.

Dr. Boivin’s laboratory (Université Laval, Quebec City, Canada) has validated this as a hMPV and typed it as G gene group 2B as per phylogram published in J General Virology 2004;85:679-686.
