The hexon typing method has been validated against classical serotyping methods with excellent agreement as reported in our recent publication:
Due to numerous requests we are making adenovirus molecular typing available to other laboratories.
Methods: Please complete an Adenovirus Typing Request Form and ship the specimen(s) on dry ice to our Center for Emerging Infectious Diseases laboratory. Shipments can be made Monday through Thursday. Upon receipt of the adenovirus positive sample, viral DNA will be extracted using the MagMax Viral RNA Isolation Kit (Ambion, Austin, TX). PCR amplification of hypervariable regions of the hexon gene (1) and fiber gene (2) will then be performed. PCR products will then be sent to the University of Iowa DNA Facility for sequencing. Sequence data will be compared to both our internal sequence database and that from GenBank. Clinical adenovirus isolates whose sequence data agrees with both the internal database and GenBank with 95% identity scores (BioEdit, Ibis Therapeutics, Carlsbad, CA) for both the hexon and fiber regions will be considered a match and the prototypic virus serotype reported via an Adenovirus Typing Report.
Turnaround time: The expected time from clinical specimen receipt to report generation is 48-72 hrs.
Costs: We have run out of grant funding so we have to have to charge a fee ($250 per isolate or specimen). Adenovirus typing requestor will be invoiced after the typing has been performed.
